FLOW CYTOMETRIC ANALYSIS IN TONGUE SQUAMOUS CELL CARCINOMA: RELATION TO DISEASE FREE SURVIVAL

Research Abstract

ABSTRACT

DNA ploidy was investigated in 19 formalin fixed-paraffin embedded blocks of tongue squamous cell carcinoma. Single cell suspensions for flow cytometric analysis were prepared according to Hedley, (1989). The DNA ploidy and histogram were calculated and compared with the incidence of tumor recurrence. DNA aneuploidy was observed in 8 (42%) tumors. The mean DNA index was 1.46 with a range of 1.74 to 1.89. Recurrence of the primary lesions was seen in ten cases, distant metastasis found in three cases while only one case showed both local recurrence and distant metastasis. The results indicate positive relationship between aneuploidy and recurrence of tongue squamous cell carcinoma

Immunohistochemical evaluation of PCNA, P53 and survivin in ameloblastoma

Research Abstract

ABSTRACT

The purpose of this study was carried out to examine the expression of PCNA, P53 and survivin in different types of ameloblastoma. The cases of ameloblastoma consisted of solid type tumors and histologic arrangements of different subtypes were observed. In some cases, more than one histologic subtype was identified in the same lesion, and each tumor was categorized according to the predominant cell pattern. PCNA and survivin immunohistochemical expression revealed stronger staining for the follicular ameloblastoma, while for p53 protein the strongest stain was detected in the plexiform type. The results indicated that the ameloblastoma has great proliferative potential, which can contribute to explain its aggressiveness and invasive characteristics

Effect of Milled Tooth Powder versus β-tricalcium phosphate as Bone Grafts on Healing of Tooth Extraction Sockets of Rabbits: An Experimental Study

Research Abstract

Abstract:

Aim: To compare the effects of milled tooth (MT) powder and β-tricalcium phosphate (β-TCP) on healing of extraction sockets in rabbits.

Methods: Sixty healthy adult male rabbits were divided into four groups (n=15). After anesthesia the right mandibular first molars were extracted and the treated materials (β-TCP or MT) were gently applied to the extraction sockets forming two groups. In the positive (+ve) control group, sockets healed spontaneously. In the negative (-ve) control group, jaws were left without any interference. Five rabbits from each group were euthanized 10, 20, and 30 days after surgery. Specimens were prepared for light microscopic examination using hematoxylin and eosin, Masson’s trichrome staining, and osteopontin. ANOVA was used to assess the statistical significance of the difference between more than two group means. Tukey's multiple comparisons and post-hoc tests were used to explore differences between the means of multiple groups.

Results: Regarding osteoid tissue formation by Masson’s trichrome staining, a highly significant difference recorded between -ve control, +ve control/10 d, β-TCP/20 d, MT/10 d, and MT/30 d groups. However, no significant differences were detected between the -ve control and +ve control, β-TCP/ 20 d, β-TCP/30 d, and MT/30 d groups. Higher expression of osteopontin were detected in the β-TCP/20 d and MT/30 d groups, and a highly significant difference was observed compared to the +ve control group.

Conclusions: MT powder is a biocompatible osteoconductive graft material that can be used as bone graft material in tooth extraction socket healing.

Histopathological Analysis and Comparison between Experimentally Fabricated Hydroxyapatite from Nile Tilapia Bone and Mineral Trioxide Aggregate as A Direct Pulp Capping Agents on Dog Pulp (In Vivo Study)

Research Abstract

Abstract

Aim: to study response of exposed dental dog pulp to the HP from Nile Tilapia bone and MTA on induction of reparative dentin formation and to inflammatory response in pulp tissue.

Material and methods: Four male mongrel dogs one year old with total of twenty teeth from each dog were selected and divided into 2 groups each group has 40 teeth according to the material used as pulp capping agents. The maxillary teeth and the mandibular teeth for each dog were capped by both materials alternatively; the cavities were then sealed by glass ionomer cement. Each group divided in 2 groups according to two-time intervals (4 weeks, 8 weeks). Animals were sacrificed and teeth were collected for histopathological analysis.

Results: specimens capped by fish bone powder after four weeks, showed non-significantly higher of inflammatory cell scores than that capped by MTA. It showed thin (score 1) to moderate (score 2) thickness of dentine bridge formation, mostly a-tubular dentin, in all specimens. After 8 weeks significant decrease in inflammatory cell infiltration scores for both groups were found, but it was significant in case of fish bone powder. Also, dentine bridge became more significant for both materials with slight histological change in the group capped by fish bone powder.

Conclusions: HA from Nile Tilapia bone could be considered as a direct pulp capping material. Also more studies needed on the fabrication of this material in order to give better result in shape and the pattern of reparative dentine formed.

EFFECT OF APIGENIN AND APIGENIN LOADED NANOPARTICLES ON SQUAMOUS CELL CARCINOMA CELL LINE (AN-IN VITRO STUDY)

Research Abstract

ABSTRACT

Background: Apigenin has effective cytotoxic anticancer properties and was used for treatment of variety of malignancies. The aim of the study is to investigate the effect of Apigenin (Apig) and apigenin loaded nanoparticles (Apig NPs) on squamous cell carcinoma cell line (HEP-2).

Material & method: Treatment of cell line (HEp-2) by different concentrations of Apig and Apig NPs was done to assess the viability of the treated cells to determine the percentages of viable cells that was monitored using MTT assay.

Results: The mean viability percentage of Apig treated cells (in relation to control cells) decreases from 103.69% to 14.41% as Apig concentration increased from 1.22 to 2500 mM with an IC50 of 51.46 mM and Apig NPs decreased the cells viability from 98.11% to 12.25% as Apig NPs concentration increased from .24 mM to 500 mM with an IC50 of .89. Morphometric analysis showed a decrease in nuclear area factor (NAF) which is an indicator of apoptosis as concentrations increased. The results were statistically significant.

Conclusion: Apig and Apig NPs have a significant anticancer effect on Hep-2 cell line

Influence of quercetin on squamous cell carcinoma cell line (HEP-2). An in-vitro study

Research Abstract

ABSTRACT

Background: Quercetin is an effective cytotoxic anticancer agent and has been used for the treatment of variety of malignancies. The aim of the current study is to investigate the influence of Quercetin on squamous cell carcinoma cell line (HEP-2) after 24 hours.

Material& methods: Treatment of cell line (HEp-2) by different concentrations of Quercetin was done to assess the viability of the treated cells to determine the percentages of viable cells that was monitored using MTT assay.

Results: the mean viability percentage of Quercetin treated cells (in relation to control cells) increased as Quercetin concentration decreased from 640 mM to 5mM with an IC50 of 200 mM. Regarding the cytotoxic effect of Quercetin and related cell cycle profile it was noticed that cell distribution showed a variable percent of arrest at different phase of cell division profile, Where there was non significant difference of arrested cells of Quercetin pre and post treatment compared with its value in non treated G0-G1 phase control cells(P>0.05) and S phase as well. While there was a significant elevated arrest of treated cells during the G2-M phase (P<0.05) and the significant difference of cell arrest at G2-M phase was type of treatment related .

Conclusion: From the results of the current study, we found that Quercetin has cytotoxic effect on squamous cell carcinoma, (HEp-2) cell line, It also induced an effect on the cell cycle distribution, and Quercetin has effect on (HEp-2) cell line resulting in apoptosis and necrosis.
