Experimental Study of The effects of Cotraceptive Steriods on Liver Proteins and Nucleic Acids In Mature Female Albino Rats

Research Abstract
Effects of administration of a wide range of doses of synthetic estrogens and progestogens on the liver total proteins, albumin and nucleic acids their role for induction of liver growth or liver impairment or as hepato mitogenic agents were studied. The study included 100 mature female albino rats, they were divided into two groups, A and B, Group A included 50 rats randomly divided into group A1: included 20 rats treated with 1.75ug Ethinyl oestradiol in 0.5 ml silicon –oil and group A2: included 20 rats treated with 7ug Ethinyl oestradiol in 0.5 ml silicon –oil in addition to 10 rats were treated by 0.5 silicon oil and represented the control group (group A3). Group B: 50 rats randomly divided into group B1: included 20 rats treated with 15ug Levonorgestrel in 0.75 ml silicon –oil and group B2: included 20 rats treated with 200ug Levonorgestrel in 0.75 ml silicon –oil in addition to 10 rats were treated by 0.75 silicon oil and represented the control group (group B3). Blood samples were used for determination of total proteins, albumin, triglycerides, alkaline phosphatase and -glutamyl transferases. Tissues used for the determination of total protein, DNA and RNA contents. The study shows that Ethinyl estradiol at 1.75ug produces significant decrease of the total protein content (P0.001), insignificant increase of the DNA and significant increase of the RNA content (P0.001). ). Mild fatty degeneration was detected in livers of 3 cases while the rest showed no abnormalities meanwhile, Ethinyl estradiol at 7ug produced insignificant increase of the total protein content, significant decrease of the DNA (P0.001) and RNA content (P0.001). Significant decrease of the total protein (P0.05) and the serum albumin (P0.001 while significant increase of the triglyceride (P0.001), -glutamyl transferase (P0.001) and alkaline phosphatase (P0.05). Meanwhile, Most of the homogenates showed fatty degeneration and cloudy swelling. Levonorgestrel at 15ug produced significant increase of the mean total protein content (P0.01), and the DNA content but insignificant increase of the RNA content. meanwhile, insignificant increase of the total protein and significant decrease of the albumin and triglyceride (P0.05 and 0.001)and significant increase of -glutamyl transferase (P0.001) and alkaline phosphatase (P0.05) All the animals showed no histopathological changes. Levonorgestrel at 200 ug produced significant increase of total protein (P0.001), insignificant increase DNA content and significant increase RNA content (P0.001). Fatty degeneration and oedema were detected. Significant increase of the total serum proteins (P0.001) but insignificant increase of serum albumin. Effects of Combined Use of Estrogenic and Progestional Steriods on Lung Proteins, DeoxyNucleic Acid and Ribonucleic Acid in Mature Female Albino Rats

Research Abstract
Objectives: Effects of administration of combination of physiological and high doses of Ethinyl estradiol and Levonorgesterel on the lung total proteins, DNA and RNA their role for induction of lung growth or as lung mutagenic agents were studied. Material and Methods: The study included 100 mature female albino rats. They were divided into two groups, I and II. Group I included 50 rats randomly divided into 40 rats treated with 1.75ug Ethinyl estradiol plus 15 ug Levonorgestrel in 0.5 silicon oil. Group II: 40 rats treated with 14ug Ethinyl estradiol plus 5mg Levonorgestrel in 0.5 ml silicon –oil in addition to 10 rats treated by 0.5 ml silicon oil and represented the control group. Lung tissues were used for the determination of total protein, DNA and RNA contents. Results: Ethinyl estradiol at 1.75ug plus 15 ug Levonorgestrel produced insignificant decrease of the total protein content, significant decrease of the DNA (P0.001) and insignificant decrease of the RNA content. Insignificant increase of total protein/DNA ratio and RNA/DNA ratio. No histopathological abnormalities were detected. Meanwhile, Ethinyl estradiol at 14ug plus 5mg Levonorgestrel produced insignificant increase of the total protein content, significant decrease of the DNA (P0.001) and insignificant increase of RNA content. Significant increase of the total protein/DNA ratio (P0.01) and the RNA/DNA ratio (P0.001) was found. The absence of any histopathological changes is suggestive that these changes are reflections of the functional performance of the lung associating the use of combined dose of estrogen and progesterone. Conclusion: Absence of any structural and vascular abnormalities upon histopathological examination could be considered as indicative that these various changes are secondary to the state of functional performance of the lung associating the use of combined dose of estrogen and progesterone. Meanwhile, both steroids seem to have no toxic or mutagenic effects in the doses used.
